ReproCELL 的 Stemgent® StemRNA™ -SR 重编程试剂盒是细胞重编程试剂盒,结合自身复制 RNA(srRNA)和 microRNA 技术,为干细胞研究者提供安全、灵活、快速、经济、高效重编程,可将来自人外周血、脐带血、人成熟和新生成纤维细胞的内皮祖细胞(EPCs)通过重编程转化为 IPS 细胞。

◆优势

来自外周血的原代 EPC 建立效率是新鲜血液和冻存的单核细胞(MNC)样品的累加。脐带血效率的数据只是来自冻存的 MNCs 。每种胶原被的 T-75 瓶最少接种 5 x 107 MNCs 。未修饰 StemRNA-SR 试剂盒步骤证明使用一个重编程实验即可得到病人之间的高效率。典型的最终结果是6孔板内每个孔有 10~20 个 iPS 细胞克隆。

Stemgent 干细胞分化试剂
自从1981年发现胚胎干细胞以来,干细胞研究工作者致力于发现培养条件和发现指导干细胞分化的主要信号分子,并且将上述成果运用到临床中。最成功的分化策略在发育生物学上有它自己的基础。干细胞研究者已经获得了体内正常发育的信号线索,在体外实验中合理设计培养条件。近来高通量机器和化学合成的发展,可以使用一些新的小分子单独或者配套添加在细胞培养基中,比如细胞因子和细胞外基质(ECM)底物。Stemgent 认为人和小鼠胚胎干细胞分化的操作流程费时费力,所以提供大量用于干细胞分化的试剂。
1. 外胚层:细胞因子(比如 SHH, Noggin),小分子化合物(比如 SHH 信号通道调节子套装)和基质(Laminin)。
2. 内胚层:细胞因子(比如 Activin A)和小分子化合物(比如 IDE-1, IDE-2)。
3. 中胚层:细胞因子(比如 BMP-4, Wnt-3a)和小分子化合物(比如 Wnt 信号通道套装)。
现在通过病毒载体工具盒可将一种成熟类型的细胞直接分化为另外一种成熟类型的细胞。受重编程成熟细胞成为诱导性多能干细胞(iPS细胞)鼓舞,Marius Wernig2 和 Douglas Melton3 找到了分化中的关键转录因子,转录因子(Ascl1, Brn2, and Myt1l)可直接分化皮肤细胞变成神经元的,转录因子(Ngn3, Pdx1, and Mafa)可将胰腺外分泌细胞分化为分泌胰岛素 beta-细胞。不逆转多能性干细胞的状态,直接重编程细胞从一种成熟细胞到另外一种成熟细胞,减少了病人组织收集和再生医疗感兴趣的细胞类型来源的实验时间。

◆产品概况
Stemgent® mRNA 重编程试剂盒与 microRNA Booster 试剂盒(00-0073)配套使用可以迅速,安全,有效制备人 iPS 细胞。该方法不发生整合,不含病毒,与临床研究相关。与慢病毒,仙台病毒和其他方法相比,16天内制备 iPS 细胞,3周内细胞可进行传代培养。慢病毒,仙台病毒和其他方法还需要花20周时间确认没有病毒残留。该系统完全免除了病毒相关的生物污染问题的顾虑。
Stemgent® mRNA 重编程试剂盒包括5种重编程因子,监测转染效率的细胞核 GFP 标记蛋白,Stemgent Pluriton™ 重编程培养基, B18R 蛋白。试剂盒和操作方法都经过充分的检测和验证。该试剂盒已经能够将人 BJ 成纤维细胞进行重编程,该细胞可作为对照。Stemgent® mRNA 重编程试剂盒可以使研究正更容易调控蛋白表达水平的时间,量化控制重编程因子。

Storage and Stability
1. Store the box (Part 1 of 2) containing the 7 vials of mRNA, the 1 vial of B18R, and the 1 vial of Pluriton Supplement at or below -70°C.
2. Store the bottle (Part 2 of 2) of Pluriton Medium at -20°C.
3. All kit components are stable for a minimum of 3 months from date of receipt, when stored as directed.
Quality Control
The Stemgent mRNA Reprogramming Kit is functionally tested for successful mRNA-based reprogramming of BJ human fibroblasts. Complete reprogramming of iPS cell colonies is confirmed by continued expression of pluripotency markers and expansion of primary iPS colonies into stable lines. All components are sterile and have tested negative for mycoplasma.
