◆泛素 – 蛋白酶体系统和癌症的功能特征

关于泛素结合和解离的TR-FRET测量引起人们高度关注。良好的FRET组合在供体和受体之间不显示重叠的光谱,高的量子产率和红移发射(例如Cy5)可以减少化合物干扰。最好的供体是由大量稀土配合物(铕或铽)与镧系元素嵌入在一个大环化合物中组成的穴状合物。它们表现出长效的荧光性,稳定性和鲁棒性,以经受不同的测定条件。
铕阴离子标记的泛素在用于测定Ub链标记或去标记时,可以作为一个理想的TR-FRET供体。Cy5标记的泛素是其理想的基于蛋白的TR-FRET配对受体。 6-FAM标记的泛素是铽标记的泛素的理想受体。
