产品概述

铁是生物体内最重要的金属元素之一,生物体内铁元素的含量虽少,但有着重要的生理作用。近年来,随着铁死亡概念的提出,细胞内铁离子的代谢过程的研究逐渐成为了研究热点。同仁化学研究所开发的 Iron Assay Kit-Colorimetric-试剂盒 是一套操作简便的比色法铁离子检测试剂盒。与其他市面上的检测试剂盒 相比 ,可以在更短的时间内进行快速检测。通过比较组织裂解液中的铁离子探针 3-(2-Pyridyl)-5,6-bis(5-sulfo-2-furyl)-1,2,4-triazine, disodium salt的 吸光度与还原成二价铁离子的三价铁标准品的吸光度,即可确定组织样品中的二价铁含量。同时,还可以通过试剂盒中的还原剂将样品中的三价铁还原成二价铁,以确定总铁含量,进而计算出样品中三价铁的含量。

实验例

组织样品中的Fe2+和 Fe3+的检测

1. 称量 100 mg肝脏组织 。

2. 加入 1 ml冷 PBS,移液器吹打 20次, Vortex振荡 10 s 14,000 RPM离心 4 min,去除上清 。

3. 将样品转移至研钵,加入液氮充分研磨成粉末后,加入 1.3 ml Assay Buffer,回收至微管中。

4. 将含有样品的微管置于超声波水浴中 5 min。(为防止样品温度上升,每 1 min间隔置于冰浴上 20 s)。

5. 16,000 g离心 10 min,除去不溶物。

6. 取 1300 μl上清液,每管分别加入 400 μl上清液,制成二价铁样品、总铁样品、样品空白。

7. Standard管 中加入 Reducer solution。 (参考图 2 H管 30 μl、 G~C管 20 μl、 B管 40 μl、 A管 20 μl)。

二价品样品管中加入 20 μl Assay Buffer。

总铁样品管中加入 20 μl Reducer solution。

样品空白管中加入 20 μl Assay Buffer。

将所有的 Standard管、样品管、样品空白管 37 培养 15 min。

8. 参考表 1和表 2,加入至 96孔板中 37 培养 60 min,检测 593 nm处的吸光度。

9. 将酶标仪数据拷贝至 “Iron Assay Kit – Excel表 ”,自动计算 出二价铁和三价铁浓度。
